If you mean to replace the check valve O-ring.
You don’t have to but, I’d change all 3 O-rings since you’re in there already.

If not, just leave that one and change the other 2 for now.
You don’t need to take the whole gun apart.
Just empty the AT(air tank) and remove and work on the gauge end.
When filling the AT back up make sure to cock the gun before filling, to take the hammer spring off of the valve stem.

No it shouldn’t be, I fill to 220 bar on my compact 177 cal all the time.
But, if you fill to fast you could burn the check valve O-ring.

Just noticed it after a recent fill. It was tuned by Ernest who did a great job. I recently filled to about 220 and wondered if that could have been an issue?
The question is, where’s the leak ?
Did you soapy water (you can use a bit of spit on it also) the 3 leak indicator holes on the AT ?
If not, I suspect it’s the check valve O-ring.
